چکیده

A macrophage-derived inhibitor of early hematopoietic progenitors (colony-forming unit-spleen, CFU-A) called stem cell inhibitor was found to be identical to macrophage inflammatory protein-la (MIP-1 a). We investigated the effect of MIP-1 a on the earliest stem cells that sustain long-term hematopoiesis in vivo in a competitive bone marrow repopulation assay. Because long-term reconstituting (LTR) stem cells are normally quiescent, an in vivo model was first developed in which they are triggered to cycle. A first 5-fluorouracil (5-FU) injection was used to eliminate later progenitors, causing the LTR stem cells, which are normally resistant to 5-FU, to enter the cell cycle and become sensitive to a second 5-FU injection administered 5 days later. Human MIP-la administered from day 0 to 7 was unable to prevent the depletion of the LTR stem cells by the second 5-FU treatment, as observed on day 7 in this model, sug-

Comparison of the Ex Vivo Expansion of UCB-Derived CD34+ in 3D DBM/MBA Scaffolds with USSC as a Feeder Layer

Objective(s): Ex vivo expansion of hematopoitic stem cells is an alternative way to increase umbilical cord blood (UCB)-CD34+ cells for bone marrow transplantation. For this purpose demineralized bone matrix (DBM) and mineralized bone allograft (MBA) as two scaffolds based on bone matrix and stem cell niche, were simultaneously used to enhance the effect of human mesenchymal pro...

Evaluation of the Effect of Ursolic Acid Compared with Ibuprofen and Dexamethasone in Synoviocyte Model for Osteoarthritis: an Ex Vivo Study

Background: Osteoarthritis is one of the most commonly diagnosed malignant diseases in the world and was previously recognized as a degenerative joint disease characterized by cartilage deformity and the formation of new bones at the edges of the joint. But this disease is not just a degenerative disease but also a biomechanical, biochemical, and cellular phenomenon. Common treatments include s...

Calcitriol modulates the effects of bone marrow-derived mesenchymal stem cells on macrophage functions

Objective(s):Some evidence showed that calcitriol has an important role in regulating growth and differentiation of mesenchymal stem cells (MSCs). However, the interaction between mesenchymal stem cells and macrophage is not clear yet.  The current study was done to investigate the in vitro effects of calcitriol on the interactions between bone marrow-derived MSCs and rat macrophages. Material...

P 141: Mesenchymal Stem Cells as Treatment in Neuroinflammatory Disease

Mesenchymal stem cells can be obtained from deferent tissues like adipose tissue, umbilical cord, placenta, skin, bone marrow, etc. These cells have regulatory effects on all types of immune cells such as dendritic cell, natural killers and lymphocytes. Mesenchymal stem cells induce inhibitory phenotypes of Antigen Presenting Cells (APCs) following their activity. They also change T cells pheno...
